Well, if you DID have the money, something like this probably costs at least a few hundred thousand dollars to start up, especially when you take into account that they are paying for the license to use the tools that Iron Lore used to make the original Titan Quest. Most of the time, whatever up front money they've used to start the studio comes from personal investment...loans, their own money, working for no pay with only the hope that they'll eventually sell enough copies to break even and hopefully earn some royalties.
Beyond start up costs, it really depends. The more money invested, the more assets they're able to produce in-house, the more work they're able to sub-contract out (I believe Titan Quest had at least one other studio that I know of that helped them out). Usually companies will trade ownership of their intellectual property in exchange for finanacing from a publisher, but that means that they lose out on a lot in the process. Putting up their own money, keeping it as small as possible means a smaller game but also means that they keep the rights to it and can do with it whatever they want.
Getting donations from people is helping them, but unless they get some sort of 'angel' investor, the game won't be a huge game....but what it WILL be is a lovingly hand crafted one that seems to have it's fans in mind and at heart so far.
How much do they need? Well, the best answer is 'Every little bit helps'

What is very good about this project is that the developers get pretty much every penny from revenues. Sure, there's distribution costs and the engine to worry about, but outside of that, you're looking at a good percentage that goes directly to the people who created the game. That's good for the, but also good for us players: It gives the developers very specific reasons to keep supporting this project and it makes them feel personally involved at every term, eg, makes for a better product!
